Computational analyses reveal spatial relationships between nuclear pore complexes and specific lamins

چکیده

Nuclear lamin isoforms form fibrous meshworks associated with nuclear pore complexes (NPCs). Using datasets prepared from subpixel and segmentation analyses of 3D–structured illumination microscopy images WT isoform knockout mouse embryo fibroblasts, we determined high precision the spatial association NPCs specific fibers. These relationships are retained in enlarged Lmna−/− Lmnb1−/− fibroblast nuclei. Cryo-ET observations reveal that filaments composing fibers contact nucleoplasmic ring NPCs. Knockdown ring-associated nucleoporin ELYS induces NPC clusters exclude A/C but include LB1 LB2 TPR or NUP153 alters arrangement Evidence number is regulated by presented. Overall results demonstrate nucleoporins act together to maintain normal organization within envelope.

Relationships at the nuclear envelope: lamins and nuclear pore complexes in animals and plants.

The nuclear envelope comprises a distinct compartment at the nuclear periphery that provides a platform for communication between the nucleus and cytoplasm. Signal transfer can proceed by multiple means. Primarily, this is by nucleocytoplasmic trafficking facilitated by NPCs (nuclear pore complexes). Recently, it has been indicated that signals can be transmitted from the cytoskeleton to the in...

Nuclear pore complexes

What do they look like? A picture is worth twice the length of this article (Figure 1). All eukaryotes have these impressive structures: vertebrates have the Cadillac model (~125 MDa) and yeast a sportier version (~50 MDa). In spite of their immense size, these structures are made of a relatively small number of proteins — about 30 — the so-called nucleoporins or nups. Defining an NPC ‘interact...

Nuclear Distributions of NUP62 and NUP214 Suggest Architectural Diversity and Spatial Patterning among Nuclear Pore Complexes

The shape of nuclei in many adherent cultured cells approximates an oblate ellipsoid, with contralateral flattened surfaces facing the culture plate or the medium. Observations of cultured cell nuclei from orthogonal perspectives revealed that nucleoporin p62 (NUP62) and nucleoporin 214 (NUP214) are differentially distributed between nuclear pore complexes on the flattened surfaces and peripher...

Pore relations: nuclear pore complexes and nucleocytoplasmic exchange.

NPCs are the sole sites of exchange between the nucleus and cytoplasm. A large family of transport factors carry cargo between the nucleus and cytoplasm through the NPC. The NPC is a huge symmetric octagonal structure comprised of dozens of NUPs. NUPs make many contacts with surrounding structures, including the NE, the cytoplasm and nuclear interior. A subset of NUPs contain repeated peptide m...
